چکیده

Processing nematodes for scanning electron microscopy requires transfer of specimens through a series of fixatives and dehydrat ion fluids (1, 2). Preferably, all steps of fixation, dehydration, and drying are performed in the same vessel to avoid repeated handling of specimens. De Grisse (2) described small chambers made of nylon mesh and glass tubing for this purpose, and Marchant (3) constructed a similar device from small polypropylene vials and Millipore R filters. In our experience, the joints between chamber walls and screens m the first design (2) were not always tight enough. Rostgaard and Christensen (4) stated stringent requirements for such a chamber and made one from brass and a luminum stock. Its construction, however, involved extensive precision machine work. This paper describes a simpler, larger variant of tile same basic design which is fabricated by modification of commonly available hardware. Threaded components allow a tight fit between screens and chamber walls, and minimize the chances of losing small specimens. T h e basic material used to construct this uni t was a standard brass compression union for 9.5-ram (3/8 inch) flexible copper tubing (Coronet Part Manufactur ing Co., 883 Elton St., Brooklyn, N.Y. 11208). Half of the threaded portions on each end of the middle piece of the fitting were cut off. T h e

A comparison of techniques useful for preparing nematodes for scanning electron microscopy.

Second-stage juveniles of Meloidogyne incognita were prepared by several different techniques for scanning electron microscopy (SEM). Sequential fixation in the cold (4-8 C) was superior to rapid fixation at room temperature, glutaraldehyde and glutaraldehyde-formalin were better fixatives than formalin alone, and critical point drying with carbon dioxide or Freon gave similar results that were...

Use of low temperature scanning electron microscopy to observe frozen hydrated specimens of nematodes.

Frozen hydrated specimens of Pratylenchus agilis and dauer larvae of Steinernema carpocapsae were observed with low-temperature field emission scanning electron microscopy. This new technique provides information about the surface features of nematodes and also allows specimens to be fractured to reveal their internal structure. Furthermore, both halves of fractured specimens can be retained, e...

A critical review towards the conception of the "Razi Style" according to the architectural analyze of Tomb and Dome Chamber in Seljuk architecture

Around the controversial subject of Seljuq architecture in Iran, the dominion of such a style in Islamic architecture, not only following the stereotype of "Razi Style"; which was proposed by Prinya, but also includes various irrelevant architectural axis. Further, considering various schools in Islamic architecture like Khurasani, Razi, Azari, and Isfahani, does not match the existence archite...

Specimen holders for simultaneous critical point drying of multiple biological specimens.

Specimen holders were developed for simultaneous critical point drying of multiple specimens of monolayer cell cultures grown on Leighton tube glass cover slips or for specimens of cells collected on silver or cellulose membrane filters. The use of these multiple specimen holders makes it possible to process several specimens in parallel and thus significantly reduce the time required for handl...

Carbon dioxide sequestration in concrete in different curing environments

This paper summarizes the results of an investigation on carbon dioxide (CO2) sequestration in concrete. Concrete mixtures were not air entrained. Concrete mixtures were made containing Class C fly ash at 0%, 18%, and 35% of total cementitious materials. Effects of three different curing environments on properties of concrete were studied: (1) moist-curing room with 100% relative humidity (RH) ...
